We found that vitamin D changes the gut microbiome boosting immune responses to cancer in mice. We used mouse diets containing different levels of vitamin D-we have no data to support any claim that SPF50 is harmful to health. For more information:

👉 TL;DR: We found that vitD ☀️ acts on the intestinal epithelium, via VDR, favouring the gut commensal B. fragilis 🦠💩 to confer systemic cancer immunity and improving immunotherapy 💊 success 🎉 (13/n)
Mice are naturally coprophagic (🍽️💩) so we thought that the transmissible tumour resistance trait might be hidden in the fecal microbiome🦠 Indeed, Gc-/- fecal transplant (FT) ➡️ WT blocked tumour growth in WT mice, whilst Abx💊 inhibited cancer resistance in Gc-/- mice 😱 (6/n)

Enter Gc globulin (Gc) 😎 Also a plasma protein, Gc cooperates with sGsn to clear circulating actin. We wondered whether Gc, like sGsn, acts as a natural barrier to cancer immunity. And alas, just as sGsn-/- , Gc-/- mice 🐭 exhibit tumour resistance 🙌 (3/n)
This phenotype is immune-dependent as it is completely lost in mice lacking T/B cells, cDC1, IFNAR or IFNGR. It also depends on the toll-like receptor/IL1R adaptor protein MyD88, typically involved in microbial recognition 🦠 And now comes the best part... 🥁 (7/n)
Just like 🐭 lacking sGsn, Gc-/- mice exhibited 📈higher activated cytotoxic T cell # in tumours, which were responsible for the tumour control. Gc loss enhances responses to T cell-based therapy💊 But this is where similarities with sGsn end and the plot thickens... 😶🌫️ (4/n)
🔎 # 1 We found that tumour resistance conferred by vitD 🍕 depends on the vitD receptor (VDR) in intestinal epithelial cells (IEC). VDRΔIEC 🐭 fed with high vitD 🍕 fail to transmit tumour control through their 💩, suggesting that vitD alters the microbiome via host cells (10/n)
🔎 # 2 With the help of

metagenomics sequencing of 💩 revealed ⬆️ B.fragilis and ⬇️ P.brevis in stools of mice with high vitD availability 🦠 Transfer of B.fragilis but not P.brevis conferred tumour resistance, which was dependent on vitD (11/n)
...whereas WT mice fed high vitD 🍕 acquire tumour resistance. Similar to Gc-/-, FT from WT mice fed with high vitD 🍕 transmits tumour control to WT mice on standard chow 😱 But how does vitD transform the microbiome to fortify 💪 cancer immunity? 🤔 (9/n)
