Our study is out @Nature . We identify non-lipid risk pathways for coronary artery disease using pooled CRISPR-interference for 2,285 genes at GWAS loci. With an amazing team we identified the genetic risk pathways for CAD that act in endothelial cells.

Thanks, Pradeep! This took a brutally long time, but we designed 2 functional assays for Corin, and tested all 64 missense variants from an exome sequencing study. The conclusions were not what we expected, but important nonetheless!

Endothelial cells are the first point of contact between cells and the blood stream, and mediate many functions related to CAD. Angiogenesis, inflammation, clotting, metabolism, and lipid transport are all EC functions relevant to heart disease.

Great work from @SebastienRubin The CAD GWAS locus at chr6p24 is called the PHACTR1 locus. However my lab and others have shown an effect on Endothelin-1 expression. This paper shows NO vascular phenotype for PHACTR1 KO. My editorial summarizing the data:

Love this paper from @JonathanBrownMD and @davidrliu . They show a single injection of a base editor extends lifespan of Progeria mice from 215 to 510 days. Kuddos for also showing that AAV resulted in liver tumors. Great promise and some perils for this therapeutic approach.

The top two genes with regulatory effects on CAD-associated genetic programs were CCM2 and TLNRD1. Unexpectedly, we found that they bind each other, and both regulate the response of endothelial cells to blood flow.

@pnatarajanmd @atvbahajournals Interesting paper. Confusing that the direction of effect is opposite of genetics (reduced PHACTR1 expression is associated with CAD risk in GWAS/eQTL). Also, the PPARg effect is unexpected because the 6p24 locus is not associated with LDL/Tgs. GWAS biology is always complicated!

Here's a new one: Ordered imaging test. Pt's insurance requires prior authorization, so I need to justify to an insurance doc. BUT the insurer is also MY HOSPITAL: Mass General/Brigham! So to order a test, my own hospital system needs me to get approval from another doc! Insane!

We found 41 CAD-associated variants that regulate CAD-associated genetic programs in our massive-scale Perturb-seq. To get here, we knocked down expression of 2,285 genes with 37,637 CRISPR guide RNAs and sequenced 214,000 individual endothelial cells. 💪 work by Gavin Schnitzler

We asked a simple question, what are the different cells that make up the aorta? Using single cell sequencing we analyzed over 6000 cells from 4 mice. With gene expression data from 1900 genes/cell we identified 10 cell types in the Aorta. (2/7)
