Out today in @eLife ! My first PD paper with @aliceyting , and my first foray into protein engineering. Rather than using optics to sculpt light, we harnessed a genetically-encoded luciferase fused to different parts of the cell’s machinery. Proximity-dependent luciferase-LOV BRET!

After a lot of characterization, we showed CaST in action by tagging psilocybin-activated neurons in the mPFC of untethered mice exhibiting psychedelic-induced headtwitch responses. We are excited for the new capabilities unlocked by CaST, and to share it with the community! 5/6

Instead, we engineered a reversible, Ca2+-dependent proximity labeling enzyme based on split-TurboID. The enzyme attaches biotin to proteins only in activated cells exhibiting elevated calcium. It’s easy to visualize/identify the tagged cells using fluorescent biotin-binders. 3/6

Existing time-gated calcium integrators require blue light – which is great when you want to capture fast time-locked dynamics, but not so great if you need to record deep in the brain/periphery without implants, or in untethered animals. 2/6

The Ca2+-dependent tagging is sensitive and fast, occurring within only 15 minutes in cultured cells. This allows you to immediately read-out activated neurons, rather than having to wait multiple hours like traditional transcription-based activity integrators. 4/6

How chemists explain things 😂 Me: But in theory, it can oxidize infinite amounts of substrate right? Lab mate: Well in theory yes. But let’s say the enzyme gets tired. Like a postdoc. In theory a postdoc can do infinite projects. But sometimes we get tired.
