The result: mesmerizing cells. You can watch transcriptional bursts in the nucleus, follow mRNA out into the cytoplasm, see which ones get translated, and watch mature protein accumulate in situ on demand.

This was fun to make: a graphic showing nucleosomes in areas enriched with H3K27ac diffuse faster than those in areas enriched with RNA Pol II (initiated form). Each image shows all tracks from a single cell. Thank you @CSUSource for promoting our work:

Collaboration work with Ashwin-san and Sasai-san at Nagoya is online . We statistically analyzed our single nucleosome imaging data , and found fast and slow nucleosomes. We can extract more and more information from our data!

O’Neil Wiggan took on this challenge by doing 2 rounds of CRISPR to track MYH9 gene expression. MYH9 encodes myosin-2a, a critical cytoskeletal motor that pushes/pulls actin so cells can move, reorganize, divide, and differentiate (cartoon adapted from Brito et al, Cells, 2020).

What it takes to publish a paper has dramatically changed over the past 2-3 decades. The 2020 expectation of exponential # figure panels required to publish a paper is getting a bit ridiculous, especially given the fact that NIH R01 modular cap of $250K hasn't changed since 1999.

At the level of translation, we see tens to hundreds of dancing polysomes, most diffusing, some directed, others localized. Sometimes we see polysomes in multi-RNA clusters that are robustly translated.

We also uncovered a hidden translational buffering system. Upon the loss of an actin depolymerizing protein cofilin, MYH9 transcription goes up, but myosin-2a translation efficiency drops by a factor of two to partially counter the response.

Cool sidenote: to find local translation sites in the midst of all the mature myosin-2a, we look for mRNA spots with FLAG signals but lacking GFP (since GFP is not yet fluorescent during translation).

For fluorescence microscopy tweets (like @rhutto3 ) who like to keep their slides with black backgrounds, here's a simple way (w/ mathematica) to invert grayscale images while leaving red, green, and blue (almost) alone. Anyone wanna make Matlab, Python, and Fiji versions?

I like this and we do need more model organisms, but not sure it's so easy (I tried daphnia once). Reminds me of a funny manual to float: 1) meditate, check; 2) feel yourself getting lighter, check; 3) float! Seriously, though, this is a great motivation:

Meandering Kyoto streets last night, managed to find this Hoko float. The Gion Matsuri festival has a 1000+ year history! The Hoko was beautifully lit up at night. Relics on the side are ancient, used to decorate the float every year.

In round one he homozygously knocked in a C-terminal GFP+MS2 tag so we could image all mature myosin-2a protein in one color and mRNA in another.

Interestingly, although we might guess the translation signals on each mRNA would get 50% dimmer, instead their signals remained pretty much unchanged, with just half as many mRNA being translated. Couldn’t see this without the single-mRNA resolution!

A notable american elm on campus. They say these trees are tetraploid, having double the number of normal chromosomes. Sad to think how many succumbed to dutch elm disease. Glad this one is here.
