Bit of a straw man... We and many others have found this and even noted on Twits. No reason to ever inject 10^13 AAV directly into brain. Dilute down to low-mid 10^12 for good expression and minimal toxicity.

@anne_churchland @sheacshl @polybiotique @cllantz @jess_cardin honestly, for anyone trained as a synaptic/cellular physiologist, the dependence of neuronal activity on the kinetics of the opsin have always been obvious, but itโ€™s really important to bring these issues to light in the systems communities.

@KordingLab I understand Sutton's point, but I think one has to first decide if you want to understand the human (or mammalian) brain, or design a computational system that can match it's performance. I appreciate both goals, but I am personally only interested in the former.

@KordingLab @shadlen The Shadlen/Movshon perspective (and also Bill and Mike's older work) doesn't include dendritic heterogeneities, where synchrony may be amplified considerably. I think computational systems neuroscience world could benefit from cell biology and membrane biophysics a bit more.

If youโ€™re interested in pushing the limits of in vivo imaging (2-photon, meso, ??) or understanding how synapses โ€œworkโ€ in the intact brain, please DM! Weโ€™re looking to hire postdocs or research scientists for this project.

@Nouronal @superkash remember that it is much more about how your grant resonates with 1-3 random people- it could get a completely different result under other circumstances- this is just the process, not a reflection of you. also, we ALL stress out anyway.
