Officially earned my PhD from
@MITdeptofBE
!
Here’s 1 lesson I learned in each year of grad school:🧵
1. Pick advisors that care about you. Not just the science, but your happiness & life goals. Your life WILL fall apart ≥1 time in grad school & you will need advisor support.
I worked at MIT, nights weekends & holidays, for 5.5 yrs as a grad. I am still working nights/wknds to finish up several projects from my PhD.
Today they emailed to inform me I was overpaid during my employment by $197 & I need to pay before they’ll confer my degree 😎
People aren’t surprised about what’s happening at Harvard, right? The PI who was inappropriate towards me during grad school won an award for “mentorship” last year. It’s rampant.
@AlexHgO
“hi, I actually checked my records and I was unpaid for approximately 3000 hours of work considering a 40 hour work week so you actually owe me $75,000. I take check, venmo, or paypal”
In full transparency I paid this and my $100 thesis submission fee (🙄) this morning, but your shared outrage made it feel less terrible🤜🤛
Onwards and upwards 💸 !

update: 6 months later, the lab is open for business 🤓
The amount of steps between signing a lab lease to actually starting the science is wild but looking around today, I can’t believe how far we’ve come. PINCH ME!!
Spent the last 8 wks working as an intern at a biotech hedge fund, very different from being at the lab bench. Top learnings I'll be applying to my PhD [1/x]:
1. Identify the key risk in any project and pursue it aggressively. If the project is going to fail, find out quickly.
Today is my 1 yr anniversary working in biotech industry!
Skill I use from PhD most frequently is SCIENCE COMMUNICATION ✍️.
I present more often in industry than grad school to diverse audiences.
Pretty slides, clear data explanations, and strong storytelling get you noticed.
I got my first invitation (me, Dr. Stopfer) to review a paper. I know most of you tenured faculty dread reviewing but for me it’s exciting to be recognized & asked!
I got engaged a few weeks ago & am frequently asked whether I’ll change my name.
No.
Why?
It’s my publications, sure, but I like my name. I’m proud of what I’ve accomplished & overcome, how I’ve grown & changed as me, Lauren Stopfer. So I think I’ll keep being her. That’s all.
6. Don’t let your definition of whether a day was “good” or “bad” depend solely on whether your experiments worked. It’s a fast spiral downwards.
In summary, you can’t do this alone, & that’s okay! There will be good months, there will be bad years. Persevere! You’ve got this!
@PamKreeger
I’m lucky to have a new job and can afford to pay them back... but $200 to a grad student living in Boston is a lot of money to have as a surprise bill!
3 years ago I lived in a basement apartment with maggots and mold. 1 year ago I lived in a dorm room with roaches, mice, and no kitchen. Now I have my PhD, my real job, and I get to live HERE 😍🤸♀️🎉!!!
It really was worth it.
*pending furniture*
2. Pick committee members following the same criteria. You will not regret this.
3. Not everyone will like you, and not everyone is meant to stay in your life forever. It okay to let people go.
4. It’s also okay to let ideas and projects go.
5. *IMPORTANT* Take time to learn about what your lab mates are working on, in detail. But also, take time to learn about who your lab mates are as people. Nobody is ever “too busy” to help someone they care about, but you have to WORK to build that relationship.
@drdanieljsharpe
Agreed! I’m fortunate that nobody made me work the extra hours- but the reality is that if I wanted to graduate sooner rather than later and have a successful PhD, 40 hrs a week just doesn’t cut it in biological research! Especially when I had TA, classes, and other duties!

day 141: Mass spec delivery! Getting a brand new MS is always exciting, but today is extra special bc this is my first experience building an MS lab solo, and completely from scratch. PINCH ME!!!
Today, our Ascend
#MS
finally came to life… ROCK ON!🤘
In <4 mo, we’ve turned a lab storage room into a top-notch MS room, with no delays/issues, despite countless teams/vendors required to finish construction & get all the pieces in place for today. I’m thrilled!

It's the beginning of a new chapter--Today I'm thrilled to announce my NEW JOB as the Head of Proteomics & Immunopeptidomics at the newly founded Aethon Therapeutics!
I am excited to build Aethon Tx's

capabilities, Stopfer-style!
Excited to begin my professional career with
@BioNTech_Group
US! I’ll be working in their protein sciences team, and can’t wait to contribute to the amazing work this company is doing 👊
This is (perhaps) an unpopular opinion, but I never viewed my PhD as a job, rather an apprenticeship that I received a stipend to live on to make ends meet. I was not rich, but I was learning from the greatest minds in class & in lab w/o taking on tuition debt (contrast to JD/MD)
Women who’ve experience an emotionally abusive relationship play over & over in their head what they’d say to their abuser if given the chance. Proud of

for saying her piece. For those of us who’ve been there…we see you.

MY SCIENTIFIC SOUL in a pre-print 🌟
Here, we show how MEK inhibition enhances tumor antigen pMHC presentation, & leverage surface density info to intelligently develop Ab-therapies against epitopes with MEKi-tunable presentation levels...
3. Communication is key. Slide design is king. Nobody cares what you have to say if you can't present it in a way that is digestable.
@BECLmit
skills really came in handy.
4. Don't spend time guessing what your supervisors want. Just ask, and adjust accordingly.
8 years ago I interviewed for a summer internship at Baxter healthcare and the recruiter told me I didn’t *look* qualified to design catheter bags. ()
Today I defend my PhD in BioEngineering at MIT ✌️

2. Go deeper. Think you've found your answer? Keep reading. The literature is rich with information to be gleaned, even if it isn't stated overtly.
3. At the same time- be efficient with lit review. Know what you are looking for, and chase the answer(s) with intention.
5. Get to know the people you work for, ask questions about their life and experiences. Even virtually, it is possible to build strong connections and create meaningful relationships.

Well, it’s finally happening! My zoom defense* is this Tuesday at 3pm EST.
Title: “Quantitative mass spectrometry-based approaches for characterizing the immunopeptidome and tyrosine phosphoproteome in cancer”
*DM me for PW if you wish to attend 🙂
1, 2, 3, MHC! Our little review on quantitative immunopeptidomics methods is now published- here we describe relative/absolute quan techniques and discuss pros/cons. Have Q’s based on your application? Reach out!
Today marks the end of my time
@BioNTech_Group
working on the proteomics team. My colleagues threw me the most EPIC farewell party- I feel lucky to have made so many friends during my 2 years here and worked on some super cool science 🦠

Science for Anyone!
I think one small way we can work to improve diversity in STEM & continue educating the public is to make the “ivory tower” of academia more accessible, so I made a 2 min vid on the science behind my recent pub!

The antigen landscape in cancer is dynamic, maluable, & easily influenced by it's environment.
By treating cancer cells with common drugs, we can manipulate both the IDENTITY & DENSITY of epitopes presented, & use those changes to our advantage💪

Lab safety classes just came in… somehow I ordered them tinted?

1st yr of PhD: maybe I’ll graduate in 4.5 yrs
4th yr of PhD: hopefully I’ll graduate before I’m a 7th yr
Me after graduation: will all my PhD papers be published by the time I turn 40???
I really wanted a job in NYC after my PhD defense so I asked the hedge fund I interned with for a job. They offered (another) internship.
**Know your worth**
My new company is incredible & as a plus their stock has doubled in the 9 wks I’ve been employed 💃
Today is 30!🎈🎂
Over the past year I’ve struggled, feeling like I’m not on the right path. Maybe the older you get, the more you know, the more there is to be insecure/unhappy about?
Flip side, there’s more to be grateful for. Today- good health & good people surrounding me ❤️
My aunt started her first dose of palbociclib today. When she googled the drug, she found my first 1st author paper in
@natcomms
about how palbo alters HLA repertoires in cancer.
Never forget why we do what we do as scientists 🎀
When I go to networking events I like to wear something BOLD so people remember if they’ve talked to me yet. Like nobody forgets if the’ve connected on LinkedIn yet with Barbie Blazer girl 🎀

Normalize balance!
Normalize fashion and makeup in lab! Normalize being hot & smart but still getting RESPECT!!!!
I am officially someone’s manager! I vow to try my best to embody & merge the best qualities in managers past and leave the toxic traits at the door. Anyone have words of wisdom?
I don’t regret my PhD weekends & late nights spent in the lab- a total dedication to research. It got me where I am today. But, I will say I’m having a LOT of fun exploring work/life balance in Cambridge and the east coast.
Heat waves definitely feel cooler at the beach! ☀️

day 122: Hardhat tour of the construction in my new mass spec room 🤓
Most startup lab spaces can’t easily accommodate the HVAC needs of a mass spec, so extensive construction is required. It’s SO exciting to see the room nearing completion!!
Sometimes it’s important to publish your results when the data doesn’t turn out how you hoped. Here’s our paper evaluating the quantitative consequences of using a protein carrier in two low-input applications: pMHC and pTyr…
New PhDs, welcome! When picking an advisor, remember what you’ll want/need from them changes over time. Hands on vs. hands off, supports career goals vs. mental heath. What doesn’t change is if they are good person. I’ve never regretted making that a priority.

In a world full of toxic academic behaviors/personalities, I feel lucky to have the advisory committee I do.
Just got off a zoom with a committee member & I’m skipping around my apt. Feels good to be supported in my science, my future goals, AND my happiness. TAKE NOTE ACADEMIA

I’ve long wondered how bad the looses are for pMHC pull downs. There are a handful of lit estimates, but I wanted to see for myself. To do this, I used our hipMHC platform and 28 isotopologue peptides shown here: (1/3)

today. I shared
one of my worst experiences was being harassed by a visiting scientist. He only called me “grad student” & said I deserved to be talked down to bc I was a 1st yr PhD.
ATTN 1ST YRS: you do NOT deserve to be talked down to.
It’s so easy to get caught up in the game of academia, wanting more papers, higher impact factors.
Today I got an email that reminded me the only “impact factor” worth caring about is the impact my work can have on human life. That’s why I am a scientist.
