10/ When we inhibited protein degradation during oocyte maturation, ELVAs retained aggregated ubiquitinated #cargos . Among such cargos degraded in ELVAs we find the aggregation-prone protein TDP-43, which has been linked to the #neurodegenerative #disease #ALS .

First, we characterised mitochondria from human and frog oocytes by proteomics and report the (first!) mitochondrial proteome in oocytes. Mitochondrial respiratory complexes were globally downregulated in early oocytes; Complex I showing a disproportionate depletion.

We discovered that early oocytes balance ROS generation and the essential mitochondrial activity by suppressing Complex I. Our discovery explains the lack of fertility issues in #LHON patients, and has implications to improve fertility in female #cancer -survivors.

5/ #EM #Tomography in collab w/ @mpicbg revealed that the aggregate-containing compartments are #clusters of endolysosomal and #autophagy vesicles tightly packed together. We therefore call them EndoLysosomal Vesicular Assemblies (ELVAs).

3/ We looked at oocytes from reproductively #young #mice . Using an aggregation probe, we found that mouse oocytes and #eggs (mature #gametes ) contain large aggregates, which disappear in embryos, suggesting that the aggregates are degraded during early embryonic #development .

8/ Second, they relocate to the oocyte cortex when the oocyte resumes #meiosis and matures into a fertilizable #egg . Unlike most endolysosomal transport in somatic cells, which relies on #microtubules , we find that ELVA relocation depends entirely on the #actin #cytoskeleton !

6/ We also find that ELVAs contain core #proteasomes , but not other #organelles like #ER , #Golgi , or #mitochondria . So ELVAs harbor both major degradative routes in the cell: the ubiquitin-proteasome system and the autophagy-lysosomal pathway.

Katsuhiko’s lab reconstituted the process of egg formation (female gametogenesis) completely in vitro - some of their reconstituted oocytes from iPSCs gave rise to fertilisable eggs which did generate live offspring!!! Consider attending his seminar if you’re in Barcelona!

Complex I is the main source of ROS in mitochondria. Biochemical assays revealed that complex I is neither active nor fully assembled in early oocytes. Thus, early oocytes become the only known animal cells where Complex I is dispensable for survival.
