The bigger the distance, the more spectral overlap between donor emission & acceptor excitation is required for a good signal.

@AliceInNanoland Both are permeating well but we’ve seen much more unspecific labeling with intracellular SNAP dyes in human cell lines. SNAP-tag was engineered from a mammalian enzyme that is also expressed in our cell line while HaloTag from a procariotic protein. So it makes sense somehow…

@LoogerL @UCSD_NIC @joachimgoedhart @LABrechaun @NathanShaner Very true! I didn’t wanna say that Halotag doesn’t mess up with protein conformation. But it allows for easy acceptor exchange without additional cloning and switching from one label to another, which would likely change the overall conformation of the engineered fusion construct

@ReidHJOlsen Related to that, I had many conversations with people claiming one assay being the best cause the delta BRET was highest. Well..different read parameters, transfection ratios and analyses often render these comparisons impossible. Plus, amplitudes are not the only factor… ➡️ #Z

@AliceInNanoland @IvanMasl0v @manorlaboratory Hey Jia! HaloTag works very well for that but we didn't have that good results with SNAP-tag. However, we were limited to use cell-permeable dyes only and that might have been the problem there. I know that SNAP-tags works very well for extracellular labeling.
