I am saddened to know my PhD committee professor, Bob Grubbs, one of the greatest chemists, 2005 chem Nobel laureate, passed away this morning. He was so humble (when I first saw him sitting on the ground listening to a talk) but serious in science and helpful in my PhD. RIP.

My second project @doudna_lab also has a topic on RNP delivery for genome editing in vivo. Different from using LNPs to package and transport gene editors, we engineered self-deliverable Cas9 RNPs through fusion to cell-penetrating peptides (CPPs).

In this project, we first used #directedevolution to engineer an efficient and ultra-stable CRISPR gene editor. The ultra-stability and desired charge density allowed the gene editor in ribonucleoprotein (RNP) format to be effectively encapsulated into lipid nanoparticles (LNPs).

Upon direct RNP injection into the mouse brain, the self-deliverable gene editors led to robust editing in vivo. We believe this self-delivery strategy would provide a facile and effective platform for genome editing both ex vivo and in vivo.

Robust gene editing upon RNP:LNP delivery was observed not only in cultured cells but also in vivo with mouse models. In particular, we developed different LNP formulations that could drive delivery specificity to the liver and the lungs for gene editing nearly exclusively.
