I will briefly summarize why I hope others find this work helpful when thinking about tripartite synapses and the role of cell adhesion molecules in astrocyte-synapse communication. 1) Astrocytes express a ton of Nrxn1, at levels roughly equal to neurons.

The Trotter lab will focus on understanding the molecular dialogue between astrocytes and neurons at “tripartite synapses” and how this communication shapes social behavior. We will have an amazing 3D-STED microscope for the microscopy geeks out there.

Another important paper that should should dampen the hype around Glia-neuron conversion. Hot take: Even if this were possible, it is hard to imagine how suddenly losing astrocyte-synapse and astrocyte-BV interactions to unspecificaly replace neurons could be a good thing.
Latest paper from our group is out, re-examining spectacular claims of single-step glia-to-neuron reprogramming. We find that genetic loss of function of Ptbp1 does not induce glia-to-neuron conversion in retina. In fact, it doesn't do much at all./1

S100beta is probably your best option since cortical astrocytes are mostly GFAP-negative. Be wary of some OPCs though with S100beta (fortunately they look much different)

Just some speculation, but I think Simona and George realize that the larger conspiracy might have been cracked and this is a last ditch effort to help facilitate an end to the OSC. I don’t think her behavior on Twitter and through the media is missed on Mueller’s team.

1) Use multichannel pipettes and strip tubes for all samples to speed up BCA and handle samples for boiling. 2) boil samples in strip tubes using thermal cycled (<60ul sample per tube). 3) use tgx gels from criterion and run at 250 V or higher (I always do 250V for 32 min)

For mouse, anti-HA.11 (purified) is the gold standard. I’ve been using this conjugated to 647 for STORM a lot recently and it works beautifully. CS has a really good / specific Rb HA mAb. Both of these are superior for me when comparing mice with Knockin HA to non-Tg.
