@biorxivpreprint I will briefly summarize why I hope others find this work helpful when thinking about tripartite synapses and the role of cell adhesion molecules in astrocyte-synapse communication. 1) Astrocytes express a ton of Nrxn1, at levels roughly equal to neurons.

@simonamangiante @ScottMStedman Also, why the sudden appeal for a pardon? And desire to support a narrative that most reasonable people will reject (i.e. Trump’a Spygate)? It seems to me that history won’t judge your actions here favorably. In fact, Mueller may already have testimony that contradicts you.

@ScottMStedman This could also be a classic false negative. Him talking to other people rather than Trump doesn’t disprove that Trump was fully aware of everything. Perhaps not in real time or it was in real time through another route (e.g. Manafort).

Another important paper that should should dampen the hype around Glia-neuron conversion. Hot take: Even if this were possible, it is hard to imagine how suddenly losing astrocyte-synapse and astrocyte-BV interactions to unspecificaly replace neurons could be a good thing.

Latest paper from our group is out, re-examining spectacular claims of single-step glia-to-neuron reprogramming. We find that genetic loss of function of Ptbp1 does not induce glia-to-neuron conversion in retina. In fact, it doesn't do much at all./1

@lyoun03 Couldn’t higher risk taking and being a pack leader alone explain higher incidence of infection? This is worded as though infection is upstream, which is great for clicks, but data doesn’t establish casaulity (i.e. T. Gondi increases risk taking and alters social hierarchy).

Using these already to great effect! For ribtotag pull-down you go from only 70% neuronal mRNA de-enrichment to nearly 100% with the miRNA targeting sequences m. Thanks for the new toolbox!

@s_meffy 1) Use multichannel pipettes and strip tubes for all samples to speed up BCA and handle samples for boiling. 2) boil samples in strip tubes using thermal cycled (<60ul sample per tube). 3) use tgx gels from criterion and run at 250 V or higher (I always do 250V for 32 min)

@christlet For mouse, anti-HA.11 (purified) is the gold standard. I’ve been using this conjugated to 647 for STORM a lot recently and it works beautifully. CS has a really good / specific Rb HA mAb. Both of these are superior for me when comparing mice with Knockin HA to non-Tg.

Really glad to finally have this preprint out! Adhesion molecules are ubiquitous at synapses but extremely diverse. So how to test how adhesion influences synapses? We made our own adhesion molecules using bacterial domains not found in vertebrates. 1/

I totally agree. @LiddelowSA addressed this at the Glia GRC a couple of years ago by calling out all of the mostly male, more senior PI’s the evening following the power hour and encouraged them to find people who attended to get the details of the discussion.

@cksalmo Slight background subtraction. Still playing with imaging parameters. Was part of a pilot last week and seems to confirm what I hoped. I think you can see a lot more microdomain events than earlier variants due to increased SNR and faster kinetics. More to come on this front.

@StefanieRobel Coming from a huge lab (usually 30+ postdocs), this is a criticism we field constantly, which is certainly unfair to trainees. A well written training plan that leverages the strengths of a lab but incorporates co-mentors to fill in gaps and assuage those sorts of critics helps

@dannybarefoot Sorry, that is not true. He has always said that he personally believes in M4A but that the American people needed to be the ones to decide as it happens by who they get their insurance from. He said that if he is right, M4A would likely win the day.
