We present rapid projection imaging under variable viewing angles. We apply this method in light-sheet and spinning disk microscopy to visualize 3D samples in real time. We further demonstrate that 3D information can be gathered from a set of projections.

This is nuts! With this optical unit, you can multiply the scan rate of a galvo (or any optical scanner) with the number of lenslets. In their work, a 16kHz resonant galvo becomes a 592khz scanner with a 37 lenslet pair! Wow!! By Jerome Mertz' group:

Cleared tissue ASLM is online: It enables isotropic, subcellular resolution over large field of views and is compatible with all clearing techniques (n=1.33-1.559). We use ctASLM to image mouse brains, kidneys and bone marrow. Image shows stem cell niche.

We present projective oblique plane structured illumination microscopy (POPSIM): POPSIM allows imaging of an entire cell under structured illumination using nine 2D projections, and as such presents a rapid alternative to conventional 3D stacking in SIM.

We present a new spin on extended depth of focusing in 2 photon microscopy. We use the phase mask form @FourierPower in reverse, and with ultrafast laser pulses instead of fluorescence: simple, high light-throughput, achromatic and pulse multiplexing.

We present a meso Oblique plane microscope. A microprism below the sample tilts the sheet, improving resolution and sectioning with a low magnification objectives. We analyzed blood flow in zebrafish embryos at 12Hz.

We are happy to present a new spin on remote focusing. Our method converts lateral scanning into axial refocusing. We demonstrate spherical-aberration-free remote focusing at a rate of 12kHz and speed up ASLM imaging by one order of magnitude.

Structured illumination with 4 beam interference by mirroring the zero order beam. Very impressive work by the lab of Hari Shroff. I did not believe this would be possible, as closing the OTF gaps appeared hard. I am very excited that they pulled it off!

The projections are computed optically, no numerical processing is needed and it goes fast. So you can watch them live on the camera stream, or also via eye-pieces.

An X-ray "light"-sheet microscope: The sheet is < 130 nm thick over a ~400 µm × 400 µm FOV  and excites nanoparticles emitting in the visible. Since LLSM already claims the term ultra-thin sheets, I am not sure what to call those. Hyper-ultra-thin?

#FluorescenceFriday Vasculature in Zebrafish, imaged with a diffractive OPM, inspired by @benjulab and Maximilian Hoffman. I like it, captures entire embryo up to 5DPF at ~1Hz, resolution good enough to see nuclei of endothelial cells in this example.

#FOM2023 I will present projective oblique plane structured illumination microscopy (POPSIM). You can project a cell into a 2D plane, and perform Gustafssonian SIM. Combined with confocal projections, you can further choose how much of the cell you project and do SIM on.
